It will be revealed in the future, as it will be explained to Serena rather soon. However, to give you a general idea, because of how successful Serena was with her career in the CIA and in SHIELD, her father wants her dead, hence why she was targeted. The reason why Ivankov said he wanted her dead, is to get a rise out of her, plus slightly reveal that her father was HYDRA all along, as he knew her father's history and why he joined HYDRA.
